The Joker Suicide Squad can be unlocked through his challenge. Otherwise, he can be obtained from his own Early Access Bundle, the Worst Heroes Ever Bundle, and as a rare drop from the Suicide Squad Daily Bonus Pack. However, obtaining him from a pack will not unlock him for direct promotions.

Passive
His passive does not include himself, meaning that in a full Suicide Squad team, he would deal 20% more damage while taking 20% less damage and not 30% for both. Furthermore, unlike the Red Son characters, it only benefits himself and not the entire Suicide Squad team; additionally, he can be more effective by gearing him with any gear(s) that can reduce damage taken (ie. Killer Croc Companion Card EVOLVED, LexCorp Set, etc) and/or increase damage dealt. Harley Quinn/Animated can be considered a useful asset to his team due to the large damage and power generation boost he receives from her. With her and a Suicide Squad teammate, he would receive a high damage boost of 50%!
His special 1's description lists its min/max damage as 13%/26% of his damage stat, prior to upgrades (the norm is 20%/40%). It starts with a kick with min/max damage as 10%/20% of his damage stat followed by a shot which have one of the four random effects, and each comes with a visual effect:
- Poison: Deals 3%-6% damage initially, then poisons the target for 10%-20% damage total over 4 seconds. Gun glows green briefly.
- Power Drain: Deals 3%-6% damage. Drains 2 bars of power (100 units) from the opponent. Gun has electric sparks around it.
- Power Gain: Gives the Joker 1.5 bars of power, at the cost of not dealing damage on the gunshot. Gun has confetti around it and shoots out a "fake" bullet that turns out to be a flag with the word "Bang!" printed on it.
- Life Drain: Deals 2 hits of 3%-6% damage each, but Life Drain only applies to one of the hits. Gun glows red briefly.
If the kick (which cannot KO) from his SP1 deals no damage (such as when opponent is at 1 HP, invulnerability or miss), and Joker gets the power gain effect, the whole special deals no damage. This refunds back power if equipped by Tantu Totem, which stacks with power gain from the special, allowing Joker to gain 2.5 bars of power with the maxed Totem.
Interactions
Please note that battles are highly situational: this section only mentions cards that have mechanics that, for any reason, are particularly effective with/against, or particularly ineffective against The Joker. Having a card that "counters" The Joker does not mean it is always a good choice.
Good With
- Suicide Squad characters.
- Deadshot/Suicide Squad: His passive allows other Suicide Squad characters to have an unblockable special, especially his powerful SP2.
- Harley Quinn/Animated: As stated above, The Joker cards on Harley's team receive +40% damage and power gen. This boosts The Joker's already strong damage stat and the power gen allows him to gain power quickly with his basic light-combo. In fact, the damage boost he gains from her is even more than his own passive.
- Catwoman/Batman Returns: She will double the poison effect from his SP1.
- Harley Quinn/Arkham Knight: She gains 3 bars of power on Joker's Knockout.
- Lord Joker/Batman Ninja: With the help of Lord Joker, Joker's Specials deal 20% Area-Effect Damage to the tagged out opponents.
Good Against
- Superman/Injustice 2, and Reverse Flash: His Power Gain on SP1 will give Joker power while consuming their passives.
- Killer Frost/Prime, and Solomon Grundy/Earth 2: The Power Gain from his SP1 is unaffected by their power dampening.
Countered By
- Power Drain characters such as Nightwing, and Lex Luthor and/or Ra's Al Ghul's Scimitar: Power Drain can prevent The Joker/Suicide Squad from using his special 1, making his passive meaningless without other Suicide Squad characters.
- Hawkgirl/Regime: She can disable special attacks on tag-in.
- Lobo/Prime, Static/Prime, Batman/Arkham Knight, and Hal Jordan/Red Lantern: Their power drain immunity will negate the power drain effect from Joker's SP1. Batman can also be immune to DOT, negating the poison effect from his SP1.
- Aquaman/Injustice 2: He can reflect back Joker's SP1 poison using his specials with double the effect.
- The Flash/Blackest Night: Blackest Night teammates will heal and gain power from his SP1 poison.
- Batman/Batman Ninja, and Shazam/New 52: They can become immune to life drain when their passives are triggered, negating the life drain effect from his SP1.

The Joker's SP1 can only knockout the opponent with the gunshot. However, if there is a bleed active on the opponent, the kick can also knockout the opponent. This also means that, if there is no bleed active, and the power gain effect occurs, this special will not be able to knockout the opponent at all.

These stats are the final damage and health levels at level 50, per Elite rank, with ALL support cards, and without any augmentations. With the 2.13 update, Elite 8, 9, and 10 have been added. Trivia
- He has the same base stats as The Flash/Earth 2.
- This version of The Joker is based on his portrayal by Jared Leto in the Suicide Squad movie that was released on August 2016.
- His artifacts are: Bracelet, Boots, Explosive, Gun and Jacket.
- His official preview on the WB Forums showed him with the same base stats as Aquaman/Injustice 2, but this was later nerfed to 1350/1350, similar to the other 2 Suicide Squad characters' nerf and Batman/Dawn of Justice, whose damage stat was nerfed from 1400 back to 1300.
- During his Challenge, the Femme Fatale Pack was brought back due to the Expert and Nightmare stages requiring Batgirl and Wonder Woman.
- He was the opponent the player would face in the tutorial (without use of Sinestro/Antimatter) during the 2.11 Suicide Squad update, and as such, the first opponent the player would have faced in the game. However, this was reverted back to its original way in the 2.12 update.
- Both him and The Joker Unhinged/Suicide Squad use the same pistol in their Super Move that was shown in their movie appearances.
- If equipped DOT gears (e.g. The Ibistick), the kick on his SP1 will apply DOT when power gain effect occurs. Otherwise the gun shot will apply DOT.
